Learning outcomes
a) Knowledge - Students with basic knowledge of organic chemistry and peptide chemistry are welcomed to participate in this laboratory practice, which will provide hands-on training in modern peptide synthesis. The course will integrate practical work of all aspects of peptide production and related methods; and will provide the background knowledge of this research field. b) Abilities - Based on the previously acquired knowledge of former organic laboratory practice, this practice will provide expertise in a special part of organic chemistry and give experience in the use of typical reagents, equipment and instruments. c) Attitude - Learning new synthetic methods and experimental techniques as well as gaining routine in the use of a typical research infrastructure. d) Autonomy and responsibility - After the practice, participants should be able to work independently on peptide synthesis, and be aware of the safety instructions of these types of laboratory work.
Course content
The laboratory practice will contain the following aspects of peptide chemistry: • Solution phase peptide coupling, synthesis of active derivatives for the coupling • Solid phase peptide synthesis (SPPS) using manual techniques • Automated peptide synthesis, the use of synthesis robots and their programming • Automated flow-based synthesis, green chemistry in peptide synthesis • Cleavage from the solid phase and cleavage of the protecting groups • Branched chained and/or cyclic peptide synthesis • Purification on preparative RP-HPLC • Analytical characterization with HPLC and mass spectrometry
Assessment method
The final evaluation will be based on the following results: two midterm and one final written examination (50%), quality of the laboratory work (30%), quality of the lab. book. (20%).
